AimPlex™ Multiplex Assays

Customizable multiplex assays read on flow cytometers
AimPlex™ multiplex assays utilize bead populations differentiated by size and fluorescence intensity levels allowing for measurements up to 24 analytes simultaneously in a single reaction.  AimPlex assays are read on a flow cytometer equipped with a 488nm laser with the maximum emission of the bead classification dye at 700 nm.
 
Similar in principle to a sandwich ELISA, each bead population conjugated with a specific capture antibody traps the protein of interest in the sample. The amount of the analyte captured is detected via a biotinylated antibody against a secondary epitope of the protein followed by a streptavidin-R-phycoerythrin treatment. Sample protein concentrations are determined by comparing the fluorescent signals to a standard curve generated from serial dilutions of a known analyte concentrations.
 
The assay procedure consists of a 60 minute antigen and capture antibody conjugated bead incubation step, a 30 minute biotinylated detection incubation step and a 20 minute streptavidin-PE incubation step.

Premixed AimPlex Panels are pre-defined multiplex panels with premixed antibody-conjugated beads, antigens and detection antibodies.  Most premixed panels are available in both 96-test and 32-test sizes.

Individual Analyte Kits (for custom-built, premixed panels)

AimPlex™ assays are offered in groups of 12-24 analytes.  All analytes in a group have a unique bead register and can be multiplexed together in any combination for easy custom configurations. When multiple Group Analyte Kits are ordered together, antibody-conjugated beads and detection antibodies are custom premixed at no additional charge.
