Protein Solubilization

PPS Silent Surfactant is a cleavable surfactant capable of disrupting cell membranes and solubilizing proteins. At low pH, PPS hydrolyzes rapidly into cleavage products with no surfactant properties and no detergent interferences. Unlike first-generation cleavable surfactants for mass spectrometry, there is no oily film or sticky pellet to complicate sample preparation.

Nucleic Acid Digestion

BaseMuncher is a non-specific endonuclease that hydrolyzes both single- and double-stranded nucleic acids (DNA and RNA) to 5-phosphorylated oligonucleotides of 1-4 bases in length. Recombinantly produced in E. coli using the Benzonase gene in a proprietary process, BaseMuncher is a highly purified homodimer of 27 kDa subunits that has exceptionally high specific activity and is completely free of protease activity. BaseMuncher is ideal for reducing viscosity during protein purification and sample preparation.

MS Trypsin Digestion

FASP Protein Digestion Kit is used to solubilize whole or fractionated protein samples in SDS, digest with trypsin and analyze the resulting peptides by mass spectrometry. Based on the Universal Sample Preparation method developed by Wisniewski, Zoubman, Nagaraj and Mann. The FFPE-FASP Protein Digestion Kit is for preparation of proteins from formalin-fixed paraffin-embedded (FFPE) tissue samples for mass spectrometry. The resulting filtrate is free of detergents, large molecules, and other substances that would interfere with mass spectrometry analysis of the proteome.

Protein Stabilization

While stability is very protein specific, NV polymers will typically protect and stabilize target proteins. NVoy technology is an alternative to the use of detergents, fusion proteins, arginine, chaperones and a range of other common additives employed to increase protein solubility and handling. The NVoy polymer increases the solubility and stability of proteins while preventing aggregation and reducing non-specific binding. NVoy polymers are linear, uncharged ~5kD carbohydrate polymers, derivatized to make them highly amphipathic, and are easily separated from the protein when they are no longer required in solution. NVoy polymers are offered as test kits (Stabil-PAC) and bulk (Polymer Pack) for routine use.

Site Specific Proteases (Fusion Protein Cleavage)

Specialized proteases for cleavage of fusion proteins: 3C-Express targets Prescission Sites and TEV-Express, an enhanced form of a catalytic fragment of the N1a cysteine protease of Tobacco etch virus (TEV), recognizes the cleavage site of Glu-Asn-Leu-Tyr-Phe-Gln-Gly, cleaving between Gln and Gly. Both proteases have GST and His tags for easy removal using either Amintra™ NiHIS or Glutathione purification resins.
